Anything you draw on a layer will overwrite whatever is on that layer. If you make another layer on top and lock the lower one it won't affect the first layer.
Put the base on the lowest layer and work from there, unless you can't use layers. for whatever reason.

Hey, /mlp/. I'm working on a request right now and was wondering. I'm starting to try coloring everything I do, but it takes for-goddamn-ever, so would you rather I just put the B&W pictures in this thread and color them in my free time, or wait even longer for the colored ones?

Post the result without coloring, then color it if it seems like it would exponentially increase the value of the /r/, or if you yourself are interested in it.
That way the /r/ can be left out of the list for some level of completion, a drawing is delivered, and then what follows after can be left up to the artist.
